چکیده

Introduction In some regions of the world viral hepatitis is a common disease that can be severe enough to result in severe decrease in liver function and death. Liver transplantation is an effective treatment. However, it involves major surgery and it also depends on the very limited availability of donor livers. The same can also be said for extensive liver resection for metastatic cancer or severe hepatic injury. The liver is an organ that can regenerate itself, given the required time and conditions. Thus, one area of research in our laboratory focuses on the use of biocompatible polymeric artificial cells containing hepatocytes and/ or bone marrow stem cells for liver regeneration.1,2 Our aim is to study whether this can allow the liver to regenerate and recover. We have shown that artificial cells containing hepatocytes can support liver function and allow recovery of rats with 90% of their liver removed. We also have shown that artificial cells can prevent immunorejection of incompatible hepatocyes without the need for immunosuppressant. However, the supply of human hepatocytes is very limited. Artificial cells can prevent immunorejection of animal hepatocytes. However, one of the functions of the hepatocytes is the synthesis of molecules like albumin and others. These foreign proteins can lead to immunological by Professor Thomas Ming Swi Chang, OC, MD, CM, PhD, FRCPC, FRS(C), Artificial Cells Containing Stem Cells for Liver Failure

Recent Technological Advances in Hepatogenic Differentiation of Stem Cells Relevant to Treatment of Liver Diseases

Liver failure, in an acute or chronic form, is a growing health problem ranking as one of the leading causes of death worldwide. Inborn errors of metabolism characterized by defects in hepatic enzymes or other proteins with metabolic functions, such as receptors or transporters accompanied with environmental factors involve etiology and presentation of liver failure. Currently, the only establi...

H2O2-Preconditioned Umbilical Cord-Derived Mesenchymal Stem Cells Ameliorate Liver Regeneration in Acute Liver Failure-Induced Mice

Introduction: Mesenchymal stem cells (MSCs) are suitable candidates for the treatment of liver diseases. However, their low survival rate limits their efficacy following transplantation. This study aimed to evaluate the therapeutic potentials of H2O2-preconditioned umbilical cord-derived MSCs (UCMSCs) on acute liver failure (ALF) in mice. Methods: UCMSCs were pre-conditioned with different con...

Autophagy-Modulated Human Bone Marrow-Derived Mesenchymal Stem Cells Accelerate Liver Restoration in Mouse Models of Acute Liver Failure

Background: Mesenchymal stem cells (MSCs) have been recently received increasing attention for cell-based therapy, especially in regenerative medicine. However, the low survival rate of these cells restricts their therapeutic applications. It is hypothesized that autophagy might play an important role in cellular homeostasis and survival. This study aims to investigate the regenerative potentia...

Intrauterine xenotransplantation of human Wharton jelly-derived mesenchymal stem cells into the liver of rabbit fetuses: A preliminary study for in vivo expression of the human liver genes

Objective(s): End-stage hepatic failure is a potentially life-threatening condition for which orthotopic liver transplantation is the only effective treatment. However, a shortage of available donor organs for transplantation each year results in the death of many patients waiting for liver transplantation. Xenotransplantation, or the transplantation of cells, tissues, or organs between differe...

Expansion of Non-Enriched Cord Blood Stem/Progenitor Cells CD34+ CD38- Using Liver Cells

Many investigators have used xenogeneic, especially murine stromal cells and fetal calf serum to maintain and expand human stem cells. The proliferation and expansion of human hematopoietic stem cells in ex vivo culture were examined with the goal of generating a suitable protocol for expanding hematopoietic stem cells for patient transplantation. Using primary fetal liver cells, we established...
